We examined the DNA integration sites of polytene chromosomes, and the integration of plasmid DNA in the chromosomes of Paramecium caudatum. The polytene chromosomes were isolated using the flow-through method. Plasmid DNA was isolated from the cells using QIAprep spin miniprep kit. The Paramecium caudatum chromosomal DNA was digested by Bal31 nuclease, and then ligated with the plasmid DNA. The ligation mixture was digested by SalI or SnaBI, separated on agarose gels, transferred onto membranes, and hybridized with the end-labeled DNA probes. We determined the sequence of the DNA-binding region in the plasmid DNA of P. caudatum. The nucleotide sequence of the binding region of the plasmid DNA was determined to be approximately 20 nucleotides upstream of the binding site of the sequence reported previously, and the sequence was identical to the Paramecium caudatum nuclear polytene repeat DNA sequence determined by Mori et al. (Mori, M., Shimizu, K., Watanabe, M., Oka, H. & Hayashizaki, T. (1990) Nucleic Acids Res., 18, 2475-2480). In the present study, the Paramecium caudatum chromosomal DNA was shown to bind to the plasmid DNA on the chromosomal DNA in vivo. The Paramecium caudatum chromosomal DNA bound to the plasmid DNA upstream of the nucleotide sequence reported by Mori et al.[Cocaine use among Brazilian adolescents].
